A pH-cycling model lab study was conducted to measure the enamel protection mode of action of Colgate® Enamel Health™ toothpaste versus ordinary fluoride toothpaste (non-antibacterial). An in vitro methodology was employed using a robotic instrument. Enamel substrates (N=6 per cell) were prepared by embedding bovine incisors in methacrylate-based resin and polishing for a fixed time period so that all samples were visually comparable and had similar levels of hardness at the beginning of the study. Prior to testing, all enamel substrates were pre-etched with 5% citric acid for 30 seconds. Half of the substrate was masked with acid resistant tape to protect the surface as a control surface.
The testing methodology alternated 1 minute toothpaste treatment periods with 2 minute acid exposure periods according to the daily sequence of T – C – C – C – C – T (T=toothpaste treatment, C=acid challenge). The acid challenge was performed with a 1% aqueous solution of citric acid (pH 3.8). Between every treatment and challenge cycle, all enamel substrates were kept in a sterile artificial saliva solution at 37 °C.
After this regimen was conducted for 5 days, the change in micro hardness was measured. The use of a programmed robot allowed for the highest accuracy in conducting the protocol over 5 days. All substrates were exposed to treatment and challenge cycle at the same temperature and time.
